An Experimental and Modeling Combined Approach in Preparative Hydrophobic Interaction Chromatography

Chromatography is a technique widely used in the purification of biopharmaceuticals, and generally consists several chromatographic steps. In this work, Hydrophobic Interaction (HIC) investigated as polishing step for therapeutic proteins. Adsorption mechanisms hydrophobic interaction chromatography are still not completely clear limited amount published data available. addition to new on adsorption isotherms some proteins (obtained both by high-throughput frontal analysis method), comparison different models proposed literature, two approaches compared work investigate HIC. The predictive approach exploits an in-house code that simulates behavior component column using model parameters found from fitting experimental data. estimation approach, other hand, commercial software which few chromatograms. validated bind-elute runs: very informative, but effort needed high; more effective, knowledge gained lower. second also applied in-development industrial process successfully resulted predicting system, allowing optimization with reduction time sample needed.
